我々の研究室では、慢性関節リウマチ(RA)の骨髄間質細胞株でプレB細胞の増殖支持能が亢進していることを見い出し(T.Kaisho
et al. J.Immunol.
149:4088-4095,1992)、RA由来の骨髄間質細胞株に高発現し、プレB細胞増殖支持能に関与する膜蛋白としてBST-1をクローニングした(T.Kaisho
et al. Proc. Natl. Acad. Sci. USA 91:5325-5329,
1994)。重症型RAの関節滑膜表層にBST-1の発現が認められること、血中可溶性BST-1の異常高値が一部の重症型RAにのみ検出されることは、BST-1のRA病態への密接な関与を示唆する。BST-1はGPI結合型細胞膜酵素で、ADPリボシルシクラーゼ活性とサイクリックADPリボース水解酵素活性を持つ。NADからADPリボシルシクラーゼによって産生されるcADPRはウニ卵の貯蔵Ca++を放出させるセカンドメッセンジャーである。マウス(m)BST-1の遺伝子クローニングの結果、BST-1は同時期にクローニングされた初期B細胞/マクロファージマーカーBP-3と同一分子であることが判明した。さらに第6回CDワークショップでC
D157と命名された。BST-1のマウス発生期における表面発現は、胎生14日の胸腺細胞、胎生16日の肝B前駆細胞から認められる。成体マウスにおけるBST-1の発現は骨髄CD45R+CD43+CD19+c-kit+のプロB細胞の分化段階に始まり、成熟B細胞で低下する。Tリンパ球系では胎生15日の胸腺細胞や成体胸腺Lin-CD4-CD8-CD3-Thy-1+のT前駆細胞中、CD44-CD25+
とCD44-CD25-
の段階に限局してBST-1の発現が認められた。したがってmBST-1はリンパ球初期発生で最も重要なIgH鎖やTCRb鎖の遺伝子再構成の時期に機能している可能性が示唆される。ヒト末梢血の顆粒球、単球、およびin
vitroで分化したマクロファージにもBST-1は発現される。骨髄単球系細胞株U937やTHP-1のBST-1を抗BST-1抗体で架橋すると130kDaの蛋白(p130)のチロシンリン酸化が誘導された。また遺伝子導入細胞株CHO-BST-1ではBST-1の架橋によりp130のチロシンリン酸化と100kDaの蛋白の脱リン酸化が誘導され、増殖が阻害された。これらの結果は、BST-1が受容体として機能し得ることを示す。本研究では、骨髄・リンパ組織の間質細胞、骨髄球系細胞、リンパ球前駆細胞といった多彩な細胞に発現するectoenzyme
BST-1の受容体としての機能を明らかにし、RAの病態生理における役割を明らかにする。そのために、1)
BST-1に会合する細胞表面分子やリガンドの解析によりBST-1の情報伝達機構を明らかにする。2)
遺伝子ターゲッテイング法により作製したBST-1欠損マウスの解析によりBST-1の血液細胞分化、免疫応答、ストレス反応における役割を明らかにする。3)すでに作製した可溶性BST-1トランスジェニックマウスを用いて可溶性BST-1の過剰発現が生体反応やRAの病態に及ぼす影響を解析する。
